好氧堆肥过程常存在分层现象,对于发酵产物的均一性和整体质量存在影响。为探究好氧堆肥物料病原菌灭活的分层情况,该研究以不翻抛(T1)为对照组,设置3组好氧堆肥试验(T2高温期翻抛、T3降温期翻抛、T4高温期和降温期翻抛),对牛粪好氧堆肥过程中常规理化性质和病原菌去除效率等进行分析,并利用克里格法(KRIGING)对堆体不同位置的温度和含水率进行插值分析。结果表明,4个处理发酵温度均达到了《畜禽粪便堆肥技术规范》要求,高温期最长的处理为T3和T4,达到7 d,T4处理的病原菌去除率最高,达到97.15%;从分层规律来看,堆体含水率从高到低为上层、中层、下层;好氧堆肥物料病原菌去除效率顺序从高到低为中层、下层、上层,4个处理病原菌去除率均不低于95.56%。综合考虑,高温期和降温期翻抛(T4处理)更有利于牛粪和秸秆混合堆肥物料整体实现无害化。该研究可为牛粪好氧堆肥过程中病原菌杀灭和翻抛参数的优化提供技术指导。  相似文献

Continuous cropping can be a serious problem in Chinese soybean production. This can result in yield reduction, root diseases, and changes in microbial community structure. We studied community structure, clone libraries, and abundance of ammonia oxidizing bacteria (AOB) in soybean fields there were in continuous soybean production for up to 17 years (SC17). Results showed that the potential nitrification rate (PNR) and amoA gene abundance of soybean in continuous cultivation for seven years (SC7) was 0.34 µg NO3? g?1 and 4.71 × 10?5 amoA gene copies/g dry soil, respectively. These values were lower than other treatments. Phylogenetic affiliation analysis based on blast results of amoA gene clone sequencing showed that the sequences belonged to seven clusters: Cluster 1, Cluster 3b, Cluster 3a.1, Cluster 3a.2, Cluster 9, Cluster 10, and Cluster 4. Correlation of AOB community compositions with environmental factors was performed using canonical correspondence analysis (CCA). Results indicated that the composition of AOB communities in maize–soybean (MS) rotation and continuous cropping of soybean for two years (SC2) were positively related to the PNR of soil, soil moisture, and soil total nitrogen content. Soybean fields continuously cropped for 11 years (SC11) and SC7 fields had AOB community compositions that were negatively related to these factors. The AOB community composition of SC17 was positively correlated to the soil total carbon content of soil. The results in this study indicate that the potential activity and abundance of AOB community in soil significantly changed after seven years continuous cropping compared to other continuous cropping intervals. Cropping systems have important effect on the diversity of functional microorganisms and associated nitrogen cycles.  相似文献

土壤中的有机污染物可从根系进入植物体内,并可进一步通过食物链富集,从而威胁人群健康。植物根际微生物种类繁多、数量巨大,其中很多根际细菌可通过成膜作用在植物根表形成细菌生物膜,协助植物抵抗外界的不良环境或促进植物生长。有机污染物在被植物根系吸收的过程中,多需经过根表细菌生物膜这一特殊界面。综述了根际细菌在植物根表的成膜作用,以及根表功能细菌生物膜对污染物根际环境过程的影响及作用机理,分析了利用根表功能细菌生物膜调控植物吸收有机污染物的可行性,试图为防治土壤有机污染、降低作物污染风险、保障农产品安全等提供理论依据。  相似文献

Long-term preservation of ammonia-oxidizing bacteria had been achieved until now by using the silica gel-freezing method. Recently, the freeze-drying method for the preservation of microorganisms has been widely used, and has been applied to the preservation of ammonia-oxidizing bacteria. The 10% skim milk-l% pyruvate or glutamate ratio was optimum as a dispersion-medium, and the preservation temperature of the freeze-dried cells was effective at -80°C. This method of preservation was found to be considerably more effective than the silica gel-freezing method.  相似文献

Aiming at learning the effects of soil conditions and cultivar on the bacterial diversity in the rhizosphere of soybean (Glycine max(L.) Merr.), bacterial communities associated with four soybean cultivars grown in two soils were revealed by terminal-restriction fragment length polymorphism (T-RFLP) combined with sequencing analysis of a 16S rDNA clone library. Lower bacteria diversity was found in soil A which has higher salinity and nutrient contents, while the highest bacterial diversity was found in the rhizosphere of cv. Jidou 12 in both soils. These results revealed that both the soil conditions and soybean cultivar affected the community composition of rhizosphere bacteria, but the effect of soil conditions was greater than that of soybean cultivar as demonstrated by the principal component analysis. It also revealed that the abundant rhizosphere bacteria may also the main symbiotic or non-symbiotic nodule endophytes.  相似文献

Abstract

The Zn content in 8 soil chemical fractions was determined for 21 greenhouse soils and for 8 reference (open field) soils from Kochi Prefecture, Japan to investigate the forms of spontaneously accumulated Zn in the greenhouse soils associated with heavy application of fertilizers and manures. Sequential extraction method was applied to every soil and each Zn fraction was designated as exchangeable (Ex-Zn), Pb-displaceable (Pb-Zn), acid soluble (Aci-Zn), Mn oxide-occluded (MnO-Zn), organically bound (OM-Zn), amorphous Fe oxide-occluded (AFeO-Zn), crystalline Fe oxide-occluded (CFeO-Zn), and residual (Res-Zn) fractions. The Zn content of the greenhouse soils was significantly higher than that of the reference soils in every fraction, except for the CFeO-Zn, and Res-Zn fractions. The Pb-Zn, Aci-Zn, and MnO-Zn fractions showed a difference of more than 60% in the total Zn content between the two soil groups. The amounts of Zn extracted in the Pb-Zn, Aci-Zn, MnO-Zn, AFeO-Zn, and CFeO-Zn fractions of the greenhouse soils increased con-comitantly with the accumulation of applied macro-nutrients. These results indicated that the accumulation of Zn in greenhouse soils caused by intensive fertilization had proceeded through specific adsorption onto or occlusion by the oxides and hydroxides of Fe and Mn in soils.  相似文献

Abstract

Dissolved humic acid (HA) and fulvic acid (FA) prepared from a Dando brown forest soil (Typic Dystrochrept) inhibited the growth of soil bacteria degrading DDT (1,1,1-trichloro-2,2-bis(4-chlorophenyl)ethane) in the culture. The population of DDT-degrading Gram-variable rod Bll6 decreased by the application of both HA and FA, suggesting the presence of bactericidal effect. Such inhibitory effect was stronger for HA and resulted in a lower degrading activity of DDT in the culture of Bll6. No inhibitory effect was observed on the growth of DDT-degrading Bacillus sp. B75. The electron spin resonance spectra showed the presence of organic free radicals in both HA and FA. The relative concentration of the radicals was higher in HA. Storage of HA solution for 3 months at 4°C decreased the concentration of the radicals as well as the inhibitory action. The addition of catalase decreased the inhibitory effect of humic acid. It is suggested that a hydroxy radical, which is derived from free radicals of humic substances, is involved in the inhibition of bacterial growth and degradation of DDT.  相似文献

ABSTRACT

Cucumbers are produced in integrated hydroponic and aquaculture systems (aquaponics). Aquaponics balances pH for plants, fish, and nitrifying bacteria. Nitrification prevents buildup of toxic waste ammonia by conversion to nitrate (NO3 ?)- nitrogen (N). The pH for hydroponic cucumbers (5.5–6.0) and nitrification (7.5–9.0) requires reconciliation to improve systems integration. Cucumbers were grown at pH of 5.0, 6.0, 7.0, and 8.0 with additional foliar sprays at pH 7.0 and 8.0. Plant shoot dry weight, length, N, and phosphorus (P) content at 14 DAT were similar from pH 5.0 to 7.0, but reduced at pH 8.0. Nutrient solution and shoot dry matter Mn and Fe decreased as pH increased. Foliar sprays had no effect on cucumber fruit yield. Early yield was higher at pH 5.0 compared to pH 8.0 but total yield was unaffected by pH. Cucumbers in recirculating culture may be maintained at pH levels more optimum for nitrification (7.5–8.0) except during production for early season markets.  相似文献

Methane-oxidizing bacteria (MOB, methanotrophs) limit the flux of methane to the atmosphere from sediments and soils, and consume atmospheric methane (King 1992; Oremland and Culbertson 1992). IPCC (1995) reported that an aerobic soil is equivalent to a sink of 10–20% of methane emissions. Hence MOB play an important role in regulating the atmospheric methane content (Mancinelli 1995). Over the last 20y, although a large amount of information has been supplied on the biochemistry of MOB, few ecological investigations have been devoted to them so far (Holzapfel-Pschorn et al. 1985; Oremland and Culbertson 1992).  相似文献
